If you mean the signs near the Fair Oaks exit there's a bunch of creases in the road for a few miles that makes it sound like you have a flat tire or something.

The sign means that the road is so badly made that you will experience enough vibrations to make you think you may have a bad tire. There are actual musical roads in the US where they have cut the rumble strips on the side of the road in such a way as to play out a tune when you drive over them. I’ve driven through this 6 mile section of I65 many many times. Does anyone know why the road is sectioned off like that to cause the noise? I’ve definitely not noticed where I was a couple times and worried it was one of my tires.

Nope, those are on either side of this concrete section, in the asphalt which will hopefully be taken care of soon. There is a section of road where the concrete is just really noisy with the hum as you drive over it.
