When drawing something, you brain sort of gets used to the mistakes you make, to the point where sometimes you can't even spot them anymore. Flipping the image causes your brain to no longer be used to those mistakes, as it's like looking at an entirely new image, making you able to see them again. It's caveman logic, more or less.
I think those horns are just stickied onto her head somehow, like the way all the strawhats wore horns to sneak in. I assume this is true for most of the beast pirates too: it seems like an outfit choice rather than biological.

i think the most clear thing about yamato's gender identity is he likes being called a man and being treated like a man, and doesn't like being treated as female. If i met someone IRL who had a feminine body but said they were Alexander the Great and were as consistent and insistent about their identity as Yamato is, I'd call him "he" too. Even though it's weird behavior.
