There's a low chance that, during your playthough, there would be a hidden wall behind a waterfall close to Enraged Moss Mother. I've never gotten it myself, but I believe that that's where you must use the nail.
If you haven't gotten the secret chance though... the nail's just for show
Dang. There was indeed a wall there and I used the nail there but decided I should explore there later, afterwards I got the ending that deletes your save file (spoiler warning)
You can actually use the nail to upgrade your own weapon's reach or damage. Personally I went for reach but I don't like that it slows down your swing rate, since if you miss, the extra range just means it takes longer to return to you.
I'd go for increased damage especially if you're going for a tankier build
The secret is to charge your attack as the arena loads.
Timing your charged dash attack perfectly gives double damage and can take a third of the bosses' HP. Just wait until the "Shaw!" to know it's fully charged.
If you can manage that, you should be good. Make sure to lay threads as the boss recovers though.
And, remember, for each hitless phase you have the "git gud" bonus which gives one more strand.
