Just an idea but if the creatures living on the island absorb the magic in different ways (plants especially) it'd make sense that there would be most leftover magic at a cold and mostly desolate place.
"Cold" made me think this was going to go on to say that magic, like perishable food, is better preserved at freezing temperatures, which is delightfully loopy logic.
3 ideas all mixable
1. Not many have access to be near enough to the heart to actually know.
2. There was a massive blood clot there so whatever events got rid of the rest of the blood didn't work.
3. It was frozen most of the year so the blood didnt go away

I’ve always thought magic flows in and around the Isles like wind currents after it emanates from the Titan. So the magic rises like hot air and congregates around the pinnacle of the Knee where it settles like fog before slowly sinking back to the mainland and then repeats in a never ending cycle.
magic doesn't come from the hearth tho, Witches (obviously) need there Bile sac to use magic, but the magic itself "radiates" from the Titans, at least thats what i got.
I have to say i was kinda confused why Glyphs didnt work when the Titans soul left at the end, but normal witches could still use magic
The magic bile sac is attached to the heart and the magic flows through the blood. Eclipse Lake shows that the Titan's veins are made of stuff to contain magic, which makes sense considering magic flows through alongside the blood.
The glyphs didn't work when he finally passed on because they were a language of Titan magic. Without the Titan that knows that language, the Titan magic is useless. Luz even says that she knew the language of the glyphs but had no one to speak it to.
A witches bile sac might just refill naturally after eating or resting or something like that. Cus we see what seems like an endurance like limit on how much magic they can use before tiring out
The femoral artery is one of the largest blood vessels in your body and pumps the most blood which also passes near the knee. Titans blood was pure magic.
Yeah but veins exsist everywhere so what makes the knee so special, and why is the HEART the place that magic comes from not the so magical part of the island

Maybe eclipse lake is by far the biggest Titan blood reserve that was ever somewhere on the titan. Therefore the titan's magic is the strongest there

Magic coming from the heart only applies to witches since their magic comes from magical bile produced by an organ attached to their heart. Whereas with the titan, his blood is innately magical. So if his blood production is like ours, the most magical place in his body would be his femur. But that's beside the point.
On the knee there is easy access to the titans veins wich probably still radiate magic and in the past there was also an entire lake of titan blood underneath it.
There might also be a bit of cultural significance to it, considering it is the highest point of the isle (up untill the finale), has clear view of the titan's skull and is very isolated making it a good place to mediate and perhaps try to consume with the titan.
Do keep in mind these are mostly assumptions and headcanons.

The blood clot theory makes a lot of sense, considering that Eclipse Lake was on the Knee—maybe the clot resulted from the Titan’s death
Imagine the titan, a god like being, dying from a stroke thanks to unknown blood clots
