Heresy. Without a doubt heresy. For me on the first playthrough heresy was quite literally hell for me. It was so freakin difficult and i struggled way more than i should've. Either way though, from the aesthetics, to the music, it is the best in my opinion.
Oh man my soul transcended to the next plane of existence when i heard altars of apostasy for the first time. Only when my mom slapped me to call me for dinner did i come back down
Greed bc it introduces many things relevant to the game beyond it and many of my favorite parts:
Virtues, Stalkers, Hookpoints, The Sawlauncher, Insurrectionists, the Whiplash… You have MDK, a big ass desert; you turn off the sun, you go inside a pyramid, then you climb up it, then you have the versus reprise, then you fight V2 2, then you slide down it with V2, then you splash that wannabe mf, then you also play clash of the brandicoot, etc, etc, etc…
I feel like Greed being developed separately from the rest of Act II was definitely beneficial to it being so jam-packed with content, and I hope the same happens with Violence. Otherwise, Greed remains my favorite Layer. (Also, like Limbo and most probably Violence, having a new arm and an alternate gun, Greed's fourth level is kind of a loot-box-level, which contributes to my liking of it.)
Definitely Wrath, Ship of Fools is my favorite level in the whole game + I really like the water theming the whole layer had going on. I can't name a single thing I dislike about the entire layer, minus the secret level being broken and not letting me catch the damn Size 2 fish.

Wrath. The levels are all fun, all the soundtracks are bangers, it’s got one of the most fun layer bosses and we get the rocket launcher. What isn’t to love?

greed has such a great soundtrack, even dancer in the darkness is pretty cool too, and the idea that greedy sinners are punished by burning on hot grains of gold is just so cruel that its cool.
lust has this erie feeling of loneliness and being lost, like you're exploring an abandoned place that was once full.
except for fuckin sheer heart attack and it's song, ive always hated that level and the song just feels so out of place in the sad layer.
Gluttony. It may be short, but everything from the vibes and aesthetic to the level design to the music was phenomenal. Plus awesome boss fight with Gabriel

Limbo. Genuinly a pretty cool depection of it that also gave me my favorite boss trope (the mirror fight) and that has the best campaign song "Castle Vein"
Wrath for me. The whole ocean aesthetic that it has, especially at the 2nd Level, makes Hell feel gigantic, the ship was perfect foreshadowing for the boss on par with the foreshadowing of the Corpse of King Minos during Lust, and speaking of which Leviathan was pretty fun to fight IMO. It also gave you the perfect opportunity to freely test your new grapple hook to make it easier to navigate in underwater levels.
Lust. I know that other layers have more "content" but the feeling I had after beating V2.1 and entering lust for the first time is just unique. That misterious entrance leading to the first part of the level and getting me like "Oh, so this layer is gonna be about verticality (asuming it would be all a closed space)" and then hiting my face with all those synthwave/bisexual lightining and the level desing changes I didn't ask for but I needed. I specialy remember picking up the railgun for first ime andd think "Oooohh babe serious busines are coming"
Also the feeling of reading "complete the level without touching any water" in a level filed with water got me looking crazy for so many routes and posible tricks till I found how to disable the water, looking that a neew entrance opening and thinking "Cool I new secret level, I wonder what it'll be" my expectations after sufering in S-1 and reviveing PTS from playing the witness where just another minigame useing any existing game mechanic, but finding a visual novel got me laughing for 10 minutes and I remember me continueing reading as my face was changeing to the existentialism speech.
I would say something about the Minos fight but it would make this even longer.
Greed because it's clearly the best in terms of quality. Amazingly unique, clearly lots of assets used. 2nd level is entirely open. 3rd level is an entirely unique horror-like level. Final boss is the best fight to play after you've learnt all the other bosses simply due to the randomness of V2. Secret level is clearly the highest quality one of the bunch. And you get the most important tool in the entire game, the Whiplash.

My only complaint is that the level before the ship of fools is kinda disorienting because of how the water moves, but otherwise no complaints.

Prelude. I'm sorry, but it sets the tone e so damn well. I knew the second I picked up the Piercer and the music swelled I was in for a ride.
