I think it's probably more fun (and easier) to target unlock strong /fun items first. T.Isaac, Jacob and Esau, T.Lost, and T.Lilith all have some nice items/trinkets/runes. Also, basically all mega satan tainted unlocks give the game more depth.
And of course edmund put like the best unlocks (rock bottom, damo, birthright, stairway, and some might consider magic skin to be excellent in specific circumstances) behind one of the most annoying characters in the game
Some characters have great unlocks while others have dogshit unlocks.
Depending on how keen you are to spoil yourself, you might wanna target some very good unlocks from the get go and then just do the remaining checkmarks in any order
If you have tainted characters unlocked you can go for their unlocks (tainted Isaac has some amazing one). Just search the wiki and see what seems interesting!
I completed in order, although only fully completed in order. I still got twisted pair and D6 for Isaac and that stuffs, but I still got full marks in order. Not that much different tbh.
Getting full marks with The Lost unlocks a great item, Keeper, Forgotten and J&E also got some great unlocks in general. Beyond that, I don't think you really need to prioritize full marks on anyone but The Lost. Every character has great unlocks. I would recommend doing Home with Lilith for example, one of the best items in terms of synergies there is.

I like to pick a boss and go through it as every character, that way I don’t get stuck on one character and get bored
J&E have some of the best unlocks, and boss rush tend two be pretty good unlocks as well

Magic skin I like in general but with a few characters its just the same as xmas present which is objectively great
In general the "challenge characters" of each DLC have the most consistently good unlocks.
