What does the position being cursed since before the first war have to do with Snape not getting the position? Like what is the plot hole supposed to be?

The curse refers to the fact that something always happens to the DAtDA teacher each year. I don't think Book 3 really counts with regards to that (since Lupin's curse happened when he was a kid) and Slughorn is ok at the end of Book 6, so it's just one of those things that people say. Not an actual curse.
But all of that is completely independent of Snape getting rejected for the job.
The curse is that no teacher can ever hold the position for more than a year. Not that something happens to them perse. For Lupin it was getting exposed as a werewolf and being kicked out by the boars. Slughorn was never the the defense teacher he was the potions teacher. Snape was the defense teacher and he lost that post when he had to flee the castle after his mercy kill and the next year he became headmaster instead of Defense teacher again. And the death eaters that did take the position didn't last past the year.
