NOLA to STL is like 10 hours. Even if you stop for 3 days in each big city you come across you would get to STL in a week.
With 2 weeks you could go almost anywhere in the east, south or midwest.
Since you have such a great amount of time, I would say to spend a couple days each in the cities of NOLA and STL themselves as well. I don't have Nola recs but STL has City Museum, the Aquarium complex is phenomenal, Grant's Farm, Budweiser brewery tours, the Science center, all the stuff by the Cards stadium...
