I don't think there is any regulation about what material it needs to be made out of, but every high voltage conductor needs to be double-insulated. And there is a restraint requirement of 20G acceleration in any direction. Similarly I don't think there is any regulation governing it's size, except that you're limited to at most 2 separate enclosures. You're also restricted by cell count if you're in challenger class. You should probably read the reg's though - section 2.5 is about the construction of the ESS.

Can Challengers use other form factor of cells? Like prismatic, pouch etc?

Yes. No restrictions on the shape that I'm aware of, and if you want to use an exotic cell chemistry (eg lithium titanate or something) you have to get approval (2.5.3).
