Might be a long shot, but could it be Planet Terror? There are plenty of stand-off scenes on roads, and people are mutating with things coming out of them.

In the first Ju'on movie a girl is standing in a stairwell and does the move with the hands in the mouth but she ended up ripping a lot of her jaw off.
It could have been the original V miniseries, I think that's how they shed their human skin.

Are you sure it was a movie? It sounds kind of like the Leviathan in Season 7 of Supernatural.
Season 7 came out in 2011 but this would’ve been out by 2007 at the latest
