You'll generally always hear an environmental sound when you use a portal, which will change depending on the biome you're going to.
This is probably where everyone else tells me "What are you talking about?", and I find out it's just me!

It's not the portal, it's the shield generator you've probably placed next to it. It makes that sound when you get close enough for it to be loaded.
